1993年到1998年先后在美国约翰-霍普金斯大学医学院神经科学系和
加州理工学院生物学部做博士后研究;1999至2007年先后在美国
德州农工大学生物科学与技术研究所任助理教授、副教授及
终身教授。
2007年受聘回国加入
华东师范大学,组建生命医学研究所并任所长;2011年12月起担任上海市调控生物学重点实验室主任,同年起担任教育部创新团队带头人、国家重大科学研究计划(973项目)首席科学家;2012年入选
上海市优秀学科带头人,2012年起任
华东师范大学生命科学学院院长。
2023年6月14日,国家自然科学基金委员会公布2023年度医学科学部基础科学中心和创新研究群体项目专业评审组组成名单,刘明耀在列。

研究领域
刘明耀教授致力于G蛋白偶联受体(GPCR)在个体发育和肿瘤及重大疾病发生发展过程中的功能及信号转导机理研究,以及靶向GPCR的新药研发。
G-蛋白偶联受体及其信号转导途径基因敲除和转基因动物。
血管新生与分子药理学。
生殖和代谢疾病。
肿瘤生长与转移的分子机理。
刘明耀教授的研究领域主要涉及发育和疾病过程中G-蛋白偶联受体(GPCRs)及其信号转导途径在胚胎发育和疾病发生过程中的作用(包括
生殖系统、癌症、
糖尿病、骨骼和心血管系统)。G-蛋白偶联受体是已知数目最多的细胞表面受体家族,它们把多种多样的胞外信号传递到细胞内,并通过和其它信号转导途径间的相互作用广泛参与细胞增殖、分化、迁移,尤其是各类生理活动和疾病的调控。刘明耀教授实验室研究的总体目标就是利用基因敲除和转基因动物技术平台研究构建疾病模型并研究GPCRs在生物体中的功能;利用
细胞信号转导网络技术平台来阐明GPCRs在正常和疾病状态下的细胞和分子机制。其最终目标是开发特异靶向性膜受体和信号途径的疾病治疗药物。研究领域集中在G蛋白偶联受体(GPCRs)及其信号转导途径,包括1)发育和代谢疾病;2)肿瘤发生,发展和转移的分子机理;3)血管新生和抗血管新生药物的分子机制及新药研发;4)炎症和肿瘤免疫中G蛋白偶联受体介导的NF-КB信号途径;5)
干细胞分化调控和人类疾病转基因动物模型的建立。

回国后作为首席科学家主持中国重大科学研究计划1项,主持国家自然科学基金重点项目2项,参与国家重大专项新药创制项目1项。
